Experiment
1. Purpose
Currently, we use many fibers. Among them, synthetic fibers occupy a lot. However, synthetic fibers often use extremely high temperatures and toxic solvents. On the other hand, silkworms can produce silk at ambient temperature using changes in salt concentration and pH. A new fiber design is expected by clarifying the silk generation mechanism of this silkworm.Silkworm silk is divided into fibroin and sericin, and this study focuses on fibroin. Fibroin consists of an N-terminal domain and repeat sequences, C-terminal domain. Fibrroin aggregates as pH decreases. It is aimed to investigate how these domains affect this aggregation mechanism.
2. Describe
The DNA of the fibroin N-terminal domain was amplified by a PCR apparatus and ligated with the vector to prepare a plasmid. It is inserted into E. coli and expressed. Measure the refined fibroin N-terminal domain under various circumstances.
3. Desired Result
I can't yet success. I hope that changes in the environment, such as pH, salt concentration, temperature and pressure of fibroin, change the structure, the state of association between proteins and inside, and thereby clarify the aggregation mechanism. Next, Since the components of the C-terminal domain and repetitive sequences have not yet been elucidated, studying these also reveals the fibrin fibrillation mechanism.
